The tingling, buzzing, numbing sensation produced by Sichuan pepper (huajiao, Zanthoxylum simulans and Z. bungeanum) and other Zanthoxylum species is caused by a class of alkylamide compounds called sanshools, principally hydroxy-alpha-sanshool. Unlike capsaicin, which activates pain-heat receptors, sanshools activate low-threshold mechanosensitive ion channels in oral sensory neurons, producing a vibration-like paresthesia at approximately 50 Hz — a tactile, not thermal, sensation. This is the má (麻, numb) component of the defining Sichuan flavor pairing má là (numb-hot).

The science

Hydroxy-alpha-sanshool (HαS) is a polyunsaturated alkylamide with an isobutylamide head group and a hydroxylated lipid tail. Its primary targets are KCNK3 and KCNK9 (TASK-1 and TASK-3), two-pore domain potassium channels (K2P channels) that set the resting membrane potential in sensory neurons. By blocking these channels, HαS depolarizes the neuron toward threshold, priming low-threshold mechanoreceptors (Aβ-fibers) for spontaneous firing. The sensation perceived is a tingling, vibrating, or buzzing paresthesia strongly resembling the 50 Hz oscillation of a vibrotactile stimulus — not pain or heat. Independently, HαS also activates TRPV1 and TRPA1 at high concentrations, contributing minor burning and pungency, but the dominant perceptual effect at typical culinary concentrations is mechanosensory. Chinese huajiao from Hanyuan county (Sichuan province) is widely regarded as the highest-quality benchmark — strong citrus fragrance, deep reddish-brown pericarp, and reliable high-sanshool content. Product labeled 'Sichuan pepper' can include Z. simulans, Z. bungeanum, or blends; Z. bungeanum from Hanyuan is the culinary gold standard. Cheaper products from other regions may deliver acceptable flavor but significantly less paresthesia.
